Improper Ferroelectric Polarisation in a Perovskite driven by Inter-site Charge Transfer and Ordering
It is of great interest to design and make materials in which ferroelectric
polarisation is coupled to other order parameters such as lattice, magnetic and
electronic instabilities. Such materials will be invaluable in next-generation
data storage devices. Recently, remarkable progress has been made in
understanding improper ferroelectric coupling mechanisms that arise from
lattice and magnetic instabilities. However, although theoretically predicted,
a compact lattice coupling between electronic and ferroelectric (polar)
instabilities has yet to be realised. Here we report detailed crystallographic
studies of a novel perovskite
HgMnMnO that is
found to exhibit a polar ground state on account of such couplings that arise
from charge and orbital ordering on both the A' and B-sites, which are
themselves driven by a highly unusual Mn-Mn inter-site charge
transfer. The inherent coupling of polar, charge, orbital and hence magnetic
degrees of freedom, make this a system of great fundamental interest, and
demonstrating ferroelectric switching in this and a host of recently reported
hybrid improper ferroelectrics remains a substantial challenge.Comment: 9 pages, 7 figure
Evaluation of the diagnostic performance of infrared imaging of the breast: a preliminary study
<p>Abstract</p> <p>Background</p> <p>The study was conducted to investigate the diagnostic performance of infrared (IR) imaging of the breast using an interpretive model derived from a scoring system.</p> <p>Methods</p> <p>The study was approved by the Institutional Review Board of our hospital. A total of 276 women (mean age = 50.8 years, SD 11.8) with suspicious findings on mammograms or ultrasound received IR imaging of the breast before excisional biopsy. The interpreting radiologists scored the lesions using a scoring system that combines five IR signs. The ROC (receiver operating characteristic) curve and AUC (area under the ROC curve) were analyzed by the univariate logistic regression model for each IR sign and an age-adjusted multivariate logistic regression model including 5 IR signs. The cut-off values and corresponding sensitivity, specificity, Youden's Index (Index = sensitivity+specificity-1), positive predictive value (PPV), negative predictive value (NPV) were estimated from the age-adjusted multivariate model. The most optimal cut-off value was determined by the one with highest Youden's Index.</p> <p>Results</p> <p>For the univariate model, the AUC of the ROC curve from five IR signs ranged from 0.557 to 0.701, and the AUC of the ROC from the age-adjusted multivariate model was 0.828. From the ROC derived from the multivariate model, the sensitivity of the most optimal cut-off value would be 72.4% with the corresponding specificity 76.6% (Youden's Index = 0.49), PPV 81.3% and NPV 66.4%.</p> <p>Conclusions</p> <p>We established an interpretive age-adjusted multivariate model for IR imaging of the breast. The cut-off values and the corresponding sensitivity and specificity can be inferred from the model in a subpopulation for diagnostic purpose.</p> <p>Trial Registration</p> <p>NCT00166998.</p
Exosomes from embryonic mesenchymal stem cells alleviate osteoarthritis through balancing synthesis and degradation of cartilage extracellular matrix
Abstract
Background
Mesenchymal stem cell therapy for osteoarthritis (OA) has been widely investigated, but the mechanisms are still unclear. Exosomes that serve as carriers of genetic information have been implicated in many diseases and are known to participate in many physiological processes. Here, we investigate the therapeutic potential of exosomes from human embryonic stem cell-induced mesenchymal stem cells (ESC-MSCs) in alleviating osteoarthritis (OA).
Methods
Exosomes were harvested from conditioned culture media of ESC-MSCs by a sequential centrifugation process. Primary mouse chondrocytes treated with interleukin 1 beta (IL-1β) were used as an in vitro model to evaluate the effects of the conditioned medium with or without exosomes and titrated doses of isolated exosomes for 48 hours, prior to immunocytochemistry or western blot analysis. Destabilization of the medial meniscus (DMM) surgery was performed on the knee joints of C57BL/6 J mice as an OA model. This was followed by intra-articular injection of either ESC-MSCs or their exosomes. Cartilage destruction and matrix degradation were evaluated with histological staining and OARSI scores at the post-surgery 8 weeks.
Results
We found that intra-articular injection of ESC-MSCs alleviated cartilage destruction and matrix degradation in the DMM model. Further in vitro studies illustrated that this effect was exerted through ESC-MSC-derived exosomes. These exosomes maintained the chondrocyte phenotype by increasing collagen type II synthesis and decreasing ADAMTS5 expression in the presence of IL-1β. Immunocytochemistry revealed colocalization of the exosomes and collagen type II-positive chondrocytes. Subsequent intra-articular injection of exosomes derived from ESC-MSCs successfully impeded cartilage destruction in the DMM model.
Conclusions
The exosomes from ESC-MSCs exert a beneficial therapeutic effect on OA by balancing the synthesis and degradation of chondrocyte extracellular matrix (ECM), which in turn provides a new target for OA drug and drug-delivery system development

Constraining Intra-cluster Gas Models with AMiBA13
Clusters of galaxies have been used extensively to determine cosmological
parameters. A major difficulty in making best use of Sunyaev-Zel'dovich (SZ)
and X-ray observations of clusters for cosmology is that using X-ray
observations it is difficult to measure the temperature distribution and
therefore determine the density distribution in individual clusters of galaxies
out to the virial radius. Observations with the new generation of SZ
instruments are a promising alternative approach. We use clusters of galaxies
drawn from high-resolution adaptive mesh refinement (AMR) cosmological
simulations to study how well we should be able to constrain the large-scale
distribution of the intra-cluster gas (ICG) in individual massive relaxed
clusters using AMiBA in its configuration with 13 1.2-m diameter dishes
(AMiBA13) along with X-ray observations. We show that non-isothermal beta
models provide a good description of the ICG in our simulated relaxed clusters.
We use simulated X-ray observations to estimate the quality of constraints on
the distribution of gas density, and simulated SZ visibilities (AMiBA13
observations) for constraints on the large-scale temperature distribution of
the ICG. We find that AMiBA13 visibilities should constrain the scale radius of
the temperature distribution to about 50% accuracy. We conclude that the
upgraded AMiBA, AMiBA13, should be a powerful instrument to constrain the

First Total Synthesis of Protoapigenone and Its Analogues as Potent Cytotoxic Agents
Protoapigenone (1), isolated from Thelypteris torresiana, previously showed significant cytotoxic activity against five human cancer cell lines. In a continued structure-activity relationship study, the first total synthesis and modification of 1 were achieved. All synthesized compounds and related intermediates were evaluated for cytotoxic activity against five human cancer cell lines, HepG2, Hep3B, MDA-MB-231, MCF-7 and A549. Among them, 24 showed 2.2-14.2 fold greater cytotoxicity than 1 and naphthyl A-ring analogs remarkably enhanced the activity
